The Protective Acations of Rutin and Quercetin in Collagen Degradation by Cu^(++) and Reactive Oxygen Species

Abstract
Flavonoids are reported to have multiple biological effects including antioxidant and metal chelating ability. Effects of rutin and quercetin on the degradation of collagen by either Cu" plus H202 or xanthine / xanthine oxidase system were investigated. Their effects on formation of hydroxyl radical and singlet oxygen were also studied.
Cu++ plus H2O2 induced collagen degradation was inhibited by oxygen radical scavengers, SOD, catalase, DMSO and DABCO. Xanthine / xanthine oxidaseinduced collagen degradation was inhibited by SOD, catalase and DABCO but not inhibited by DMSO. Rutin and quercetin markedly prevented collagen degradation by Cu++ plus H2O2. Rutin prevented collagen degradation by xanthine
xanthine oxidase but quercetin did not affect. Hydroxylation of sodium salicylate by either interaction of Cu` plus H2O2 or xanthine / xanthine oxidase was inhibited by rutin, quercetin and DMSO. Oxidation of DPBF by U. V. irradiation was inhibited by rutin and DABA, a quencher of ¢¥02 and slightly inhibited by quercetin.
These results suggest that the protective actions of rutin and quercetin on collagen degradation by either Cu` plus H202 or xanthine / xanthine oxidase may be ascribed to their scavenging action on reactive oxygen species, particulary ¢¥O2 and their effective chelating ability to Cu++
